Experimental Objective
To quantify the relative or absolute abundance of target RNA in samples such as cells and tissues, for gene expression analysis, treatment response studies, and biomarker validation.
Scope of Application
- Samples: Cultured cells, fresh or frozen tissues, blood, FFPE, bodily fluids, or purified RNA

Standard Workflow
- Determine samples, target genes, and quantification strategy
- Extract total RNA and remove genomic DNA
- Assess RNA concentration, purity, and integrity
- Normalize RNA input and perform reverse transcription
- Prepare qPCR reaction mixture and include all controls
- Check amplification curves, melting curves, and amplification efficiency
- Complete normalization, statistical analysis, and results reporting

Required Controls and QC
- No-template control (NTC), no-reverse transcription control (-RT), positive control, and stable internal reference
- Technical replicates show consistent Ct values, and dye-based melting curves show a single main peak
- Amplification efficiency and standard curve linearity meet laboratory preset ranges
Common Problems
If there is no amplification, check RNA/cDNA quality, primers, and reaction components; if replicate variability is high, improve pipetting, mixing, and bubble control; if NTC is positive, replace reagents and perform segregated cleaning.
